High-fructose diet: NAFLD, hyperuricemia and metabolic syndrome Acquired / not inherited
Defect / target: Fructose bypasses PFK-1 regulation (fructokinase/aldolase B entry)
Mechanism: Unregulated glycolytic flux generates excess acetyl-CoA → de novo lipogenesis and VLDL export, while fructokinase ATP consumption drives AMP degradation to uric acid.
↑ Accumulates: triglycerides (liver, VLDL), uric acid· ↓ Deficient: ATP (transient hepatic)
Presentation: non-alcoholic fatty liver disease, insulin resistance, metabolic syndrome, gout risk, dyslipidemia with sugary beverages/HFCS
